Pfizer Bails from Eye Alliance, Hands Glaucoma Drug to NiCox

Almost a full year after Pfizer Inc. decided to discontinue development of PF-03187207, it finally handed the compound back to its erstwhile partner, NiCox SA, and has withdrawn from its strategic alliance in ophthalmology with the company.

Financial terms were not disclosed, but NiCox, of Sophia Antipolis, France, will pay New York-based Pfizer two milestones, which are linked to the compound gaining approval in the European Union, the U.S. and Japan, and to it passing a pre-defined sales threshold. Pfizer would also receive royalties in eventual product sales.

In return, NiCox is regaining full development and commercialization rights to PF-03187207, a nitric oxide-donating prostaglandin analogue that has completed two Phase II trials in glaucoma. It also is getting back a series of research-stage nitric oxide-donating compounds in development for diabetic retinopathy and glaucoma. It will gain access to certain proprietary information on Pfizer's blockbuster glaucoma drug latanoprost (Xalatan) to aid in future regulatory filings.

The agreement has taken some time to complete. Pfizer said last August that it would discontinue development of PF-03187207 in Asia, following its failure to reach the primary endpoint in a Phase II trial in Japan. In May, it decided to cease development of the compound outside of Asia, after PF-03187207 failed to reach statistical significance in a U.S. Phase II study. "Where they put the bar was very high - higher than we achieved," NiCox CEO Michele Garufi told BioWorld International. At high doses, PF-03187207 did demonstrate an improvement over latanoprost in its ability to lower intraocular pressure (IOP), however. "Our compound has a more prolonged and sustained activity on IOP - apparently," Garufi said. "I feel confident that it still has potential market share."

The same drug mechanism underpins a licensing deal completed in April between Santen Pharmaceutical Co. Ltd., of Osaka, Japan, and Merck & Co., of Whitehouse Station, N.J. That agreement concerns tafluprost, a prostaglandin analogue that already has gained approval in several EU member states and in Japan.

NiCox plans to move PF-03187207 into Phase III studies directly "We want to push it forward - it's a drug that has a complete Phase II package," Garufi said. The Phase III program is likely to begin next year. The company has not yet decided whether to find another partner for the compound or to undertake late-stage studies itself.

That decision may well depend on what happens with its lead drug candidate. Naproxcinod, a first-in-class nitric oxide-donating cyclooxygenase inhibitor has completed a Phase III program in osteoarthritis, and an NDA filing is "on track" for the third quarter, Garufi said, with an EU filing to follow before year-end.

The compound remains unpartnered as yet. "It's taking a bit longer than we expected" he said. AstraZeneca plc originally in-licensed the compound for pain, but terminated the agreement in 2003.

NiCox entered two deals with Pfizer, in 2004 and 2006, which were worth up to €37 million (US$52.3 million) and €323 million, respectively. The company earned about €27 million in total from the alliance, Garufi said. A cardiovascular drug development alliance with Merck, worth potentially €288.2 million, is ongoing.

NiCox's stock (PARIS:COX) rose by almost 3 percent Aug. 6, when the Pfizer agreement was disclosed. It ended the day at €8.77, up 24 cents. The company reported €77 million in cash at June 30 and is funded through the end of 2010.

En retour,
Nicox est en train de développement et les droits de commercialisation
de PF-03187207, un don d'oxyde nitrique analogue de prostaglandine, qui
a terminé deux essais de phase II dans le glaucome.

Il est aussi le retour d'une série d'étapes de recherche donneurs
d'oxyde nitrique dans le développement de composés pour la rétinopathie
diabétique et le glaucome. Il permettra d'avoir accès à certains

réussi à atteindre la signification statistique des États-Unis dans une

La société n'a pas encore décidé s'il allait trouver un autre
partenaire pour la concession ou à la fin de l'étape des études
lui-même.

Cette décision dépendra de mai et ce qui arrive avec son premier candidat-médicament. Naproxcinod, une première en classe
donneurs d'oxyde nitrique inhibiteurs de la cyclooxygénase a terminé un
programme de Phase III dans l'arthrose, et d'un dépôt NDA est «sur la
bonne voie» pour le troisième trimestre, Garufi a dit, avec un dépôt de
l'UE à suivre avant la fin de l'année .
Le complexe reste unpartnered encore. «C'est en prenant un peu plus longtemps que nous attendions" at-il dit. AstraZeneca plc initialement sous licence le composé de la douleur, mais mis fin à l'accord en 2003.
Nicox a inscrit deux traite de Pfizer, en 2004 et 2006, qui ont été à
hauteur de € 37 millions (US $ 52,3 millions) et € 323 millions,
respectivement. La société a gagné environ € 27 millions au total de l'alliance, Garufi dit.Une mise au point de médicaments cardiovasculaires alliance avec Merck,
d'une valeur de € 288,2 millions potentiellement, est en cours.


Nicox du stock (PARIS: COX) a augmenté de près de 3 pour cent 6 août, lorsque la convention a été divulgué Pfizer. Il a terminé la journée à 8,77 €, en hausse de 24 cents. L'entreprise a déclaré € 77 millions en espèces au 30 Juin et est financé par la fin de 2010.

Les résultats du PF 207 sont un poil meilleur que le XALATAN (et encore.. non statistiquement significatif), équivalent à peu de chose prés au XALACOM.

Or bientôt arrivera sur le marché des médicaments qui font mieux que le XALATAN ainsi que le XALACOM (certains sont déjà sur le marché).

Il s'agit notamment du TRAVOPROST alias DUOTRAV (pas de jeu de mot merci), du BIMATOPROST, du COMBIGAN etc (désolé mais je suis certain que je l'apprends a 99% du forum, vu leur analyse concernant le PF 207)

Tout ces médicaments font mieux que XALACOM et bien sûr par transitivité vous comprendrez tout seul comme des grands qu'ils font mieux que le PF 207.
